Gleeson Score
From my MRI, it was disclosed that I had two lesions. Lesion 1 - PI-RADS Assessment Category: 4 and lesion 2- PI-RADS Assessment Category: 3. To determine the Gleeson score, are the two category scores added together?

Gleason score is from biopsy, not the MRI. As stated earlier, only biopsy confirms cancer, MRI is not confirmation. Legion could be benign.
Biopsy is graded and they look at the two most prevalent cancer in the samples.
Waiting is hard part but, need biopsy to know if you even have cancer.

There is no good reason to hold off a trip, waiting for the biopsy results. If the results confirm prostate cancer (PCa) is present, any treatment protocol won't begin immediately. PCa is slow growing, so you will have plenty of time to review and discuss treatment options with your medical team, and select the appropriate treatment option.

As Joseph said no good reason to hold off on a trip now certainly not before the biopsy. You will need to take it easy after the biopsy for a week, but that is just taking it easy not doing anything physically exerting. Even after the biopsy it would likely be several months if cancer detected before you would settle on a treatment plan, that is even if cancer were detected that needed treatment and not surveillance. Again do your trip.
BE SURE the biopsy will be an MRI fusion directed biopsy. This ensures the Urologist gets several specimen cores from the lesions identified before doing the remaining grid core samples. Good luck!
